In this study, the effects of adding sodium tripolyphosphate during the extrusion of textured wheat protein (TWP)-based meat analogs were investigated. Five TWPs (TWP-C0, TWP-C0.10, TWP-C0.25, TWP-C0.50, and TWP-C0.75) were prepared with sodium tripolyphosphate concentrations of 0%, 0.10%, 0.25%, 0.50%, and 0.75%, respectively. The fibrous structure of TWPs was analyzed by determining their textural properties, degree of texturization, microstructure, and protein bonds. When the concentration of sodium tripolyphosphate increased from 0% to 0.75%, the fibers in TWPs became more regular and finer with smaller pores, the degree of texturization increased from 2.10 ± 0.09 to 2.73 ± 0.07, and the proportions of solubilized protein from the breaking of hydrophobic bonds and disulfide bonds increased from 2.06 ± 0.14% and 1.38 ± 0.11% to 3.42 ± 0.12% and 1.74 ± 0.05%, respectively. The results of particle size, soluble nitrogen content, and free amino acids of samples during digestion indicated that the disintegration rate and protein digestibility of TWPs increased with the increase in the concentration of sodium tripolyphosphate. After gastrointestinal digestion, the total free amino acids released in TWP-C0, TWP-C0.10, TWP-C0.25, TWP-C0.50, and TWP-C0.75 were 391.5 ± 2.2, 403.9 ± 1.5, 430.0 ± 3.6, 473.8 ± 2.9 and 485.3 ± 5.73 mg/10 g digesta, respectively. Sodium tripolyphosphate may improve the protein digestibility of TWPs by forming a finer fibrous structure with a more unfolded protein structure and more hydrophobic groups being exposed to enzymes.

During plant vascular development, xylem tracheary elements (TEs) form water-conducting, empty pipes by genetically regulated cell death. Cell death is prevented from spreading to non-TEs by unidentified intercellular mechanisms, downstream of METACASPASE9 (MC9)-mediated regulation of autophagy in TEs. Here, we identified differentially abundant extracellular peptides in vascular-differentiating wild-type and MC9-down-regulated Arabidopsis cell suspensions. A peptide named Kratos rescued the abnormally high ectopic non-TE death resulting from either MC9 knockout or TE-specific overexpression of the ATG5 autophagy protein during experimentally induced vascular differentiation in Arabidopsis cotyledons. Kratos also reduced cell death following mechanical damage and extracellular ROS production in Arabidopsis leaves. Stress-induced but not vascular non-TE cell death was enhanced by another identified peptide, named Bia. Bia is therefore reminiscent of several known plant cell death-inducing peptides acting as damage-associated molecular patterns. In contrast, Kratos plays a novel extracellular cell survival role in the context of development and during stress response.

The influence of shear flow on aggregation and disaggregation in aqueous solutions of the thermoresponsive methoxy-poly(ethylene glycol)-block-poly(N-isopropylacrylamide) (MPEG53-b-PNIPAAM113) copolymer that exhibits a lower critical solution temperature was investigated with the aid of turbidity, shear viscosity, and rheo small angle light scattering (rheo-SALS) methods. The turbidity results at quiescent conditions revealed a novel transition peak in the turbidity curve at intermediate temperatures, which reflects the delicate interplay between temperature-induced aggregation and shrinking of the species. A similar anomalous transition peak (located at the same temperature) was observed in the steady shear viscosity measurements at intermediate temperatures, and the amplitude of the peak was reduced with increasing shear rate as a consequence of breakup of interaggregate chains. At low temperatures (low sticking probability), enhanced shear rate generated interpolymer aggregates; whereas in the high-temperature domain (high sticking probability) association structures were broken up as the shear rate was increased. The rheo-SALS experiments disclosed growth of aggregates at low temperatures and destruction of association complexes at high temperatures. An increase of the cloud point temperature with rising shear rate is reported, which is interpreted as being a disruption of clusters under the influence of shear stresses.

Self-association behavior of polypeptide graft copolymer and its mixture with polypeptide block copolymer and drug carrier capability of the formed micelles was examined. The results gained through fluorescence spectroscopy, transmission electron microscopy and nuclear magnetic resonance spectroscopy revealed that both polypeptide graft copolymer and its mixture with polypeptide block copolymer can self-assemble to form polymeric micelles in aqueous media. The molecular structure of the graft copolymer and blending the graft with block copolymer exert marked effects on the critical micelle concentration and the shape of formed micelles. It was found that the hydrophobic inner core of the micelles formed either by graft copolymer or mixture of graft and block copolymers can act as an incorporation site for the hydrophobic drugs. The drug loading content of the graft copolymer micelles tends to be larger when the content of the polypeptide segments in the copolymer increases. The results obtained from the drug-release studies showed that the drug-release rates are dependent on the chemical nature of the graft copolymer, the composition of the graft and block copolymer mixture, and also the pH value of the release media.
